Mechanism

  • L-5 MTHF Calcium: Active folate, reduces homocysteine, supports neurotransmitter production.

Pharmacokinetics

  • Absorption: SAMe is absorbed in the small intestine; bioavailability improves with enteric-coated formulations. Methylcobalamin and L-5 MTHF Calcium are rapidly absorbed, bypassing conversion steps required by inactive forms.
  • Distribution: SAMe is widely distributed in hepatic tissue and crosses the blood-brain barrier. Methylcobalamin circulates bound to transcobalamin II, reaching nervous tissue. L-5 MTHF Calcium distributes to plasma and cerebrospinal fluid, supporting methylation pathways.
  • Metabolism: SAMe undergoes hepatic metabolism via transmethylation and transsulfuration pathways. Methylcobalamin acts as a coenzyme in methionine synthase reactions. L-5 MTHF Calcium directly participates in homocysteine remethylation without requiring enzymatic conversion.
  • Elimination: SAMe metabolites are excreted via urine and bile. Methylcobalamin is primarily eliminated renally. Folate derivatives are excreted in urine, with some biliary recycling.
